Fenerbahce claims that Jose Mourinho's comments were taken out of context after Galatasaray accused him of racism.
Following Monday's 0-0 draw between the two clubs, Turkish Süper Lig leaders announced plans to file criminal charges against Mourinho for making "derogatory statements" about Turkish people.
The club also stated that it would file complaints with UEFA and FIFA after the 62-year-old claimed that the Galatasaray bench was "jumping like monkeys" following an early challenge and that the match would have been a disaster if a Turkish referee had been in charge.
Slovenian Slavko Vincic refereed the Istanbul derby after both clubs requested that a foreign match official take charge.
Galatasaray issued a statement calling Mourinho's behavior "reprehensible," adding that he "has persistently issued derogatory statements directed towards the Turkish people" since taking over Fenerbahce in June 2024.
"A statement made by our Technical Director Jose Mourinho after the match was completely taken out of context and an attempt was made to deliberately distort it," the club claimed.
"As every sensible person can see and understand, Jose Mourinho's statements describing the opposing team's technical staff's excessive reaction to referee decisions during the match have nothing to do with racism.
"Trying to portray this statement as racist is a completely malicious tactic. We would like to inform the public that we will exercise our legal rights in response to this pathetic slander in order to remove the competition from the field, change the agenda, and manipulate it.
Mourinho was given a one-game suspension and fined nearly £15,000 earlier this season for his reaction to Fenerbahce's 3-2 Süper Lig victory over Trabzonspor.
The Portuguese was charged with "unsportsmanlike behavior towards opposing team fans" and "diminishing the value of Turkish football and discrediting the impartiality of referees and other match officials" during an outspoken post-match interview.
